Im still 100% sold on the cell-mediated tolerance idea, in all but the most severe of cases anyway. Il-10 mimicking/overproduction & IFN esp g blockers/antagonists can put a SERIOUS dent in cellular immunity. Enough to cause huge suppresion of Th1 cell production and even a total loss of oral tolerance. Combine that with altered antigen presentation due to downregulated dendritic cell production, a proliferation of Th2 presenting innate cells due to a long term prescence of Th2 cytokines, mhc class 2 downregulation and (big one) intracellular glutathione depletion and then an altered redox state and what comes with that, I can definately see how a person would not produce a weal.
Have there been any studies done that cultured cells from a “tolerant” person, tested them particularly in the absence of Th2 cytokine/ cytokine producing cells and presented them with antigen then measured cytokine production and cytotoxic cd8 activity? Or any broad studies done for other co-infections? Or a measure of dendritic cell counts?
